I recently bought a 2013 Honda CRV, my third CRV. A week after I bought the 2013, I lost the spare set of keys. What is the best means to get a replacement key? Is a dealer the best way? Thanks

Try a locksmith first, you will probably save money. Be sure, though, that they can reset the key because they have anti-theft chips inside.

Not all locksmiths are alike - I had an ignition key issue with my '07 Ridgeline once and the good folks at the Ridge' forum recommended a 'vehicle' locksmith...Made a few calls and found a really great one.
The Dealership wanted $500+ to change out my ignition lock and give me a new key. The doors were to be operated by the old key... The vehicle locksmith rebuilt the ignition for $80. A WAYYYY better outcome.
